It would depend on what type of seat post/clamp you have. What kind of bike is it? I know that on my Cannondale Synapse, it is very tricky to adjust saddle tilt. From all outward appearances, it seemed like I could not tilt my saddle either, but a quick call to my LBS where I bought the bike resolved that. I too, have only 1 screw on my clamp...so in order to adjust tilt, it requires prying off 2 plastic "end caps" and then adjusting the tilt on each side of the post separately...then pop the end caps back on. It's time-consuming and requires a lot of patience...which is why I had my fitter take care of it during my bike fit.
